Targeted Protein Degrader Safety

Mission Statement

The collaborative mission of the committee is to advance efficient and effective translational safety assessment to maximize the therapeutic potential of targeted protein degraders for patients, by an international network of multi-partite, multi-stakeholder experts.

Working Groups

  • Cereblon-related Safety Challenges

    This group is exploring challenges related to cereblon-mediated protein degradation and mapping current tools available to evaluate those challenges, such as teratogenicity.

    Review publication: Safety considerations for cereblon-recruiting targeted protein degraders, Nature Reviews Drug Discovery, 2026

    Ongoing experimental program: Evaluating in vitro platforms to assess teratogenicity of TPDs

    Research Objectives:

    • Enhance understanding of how to interpret data generated across different in vitro platforms for teratogenicity screening.
    • Assess phenotypic effects of cereblon binders that lack degradation activity.
    • Explore targets linked to teratogenicity in the context of the in vitro assays.
    • Correlate degradation of the substrates with the readouts of the in vitro assays.

  • Study Design Challenges

    This group is discussing assays and models used to evaluate safety and highlighting specific differences for heterobifunctional degraders and molecular glue degraders compared to traditional small molecules.

    Manuscript in preparation: Preclinical safety assessment of Targeted Protein Degraders: Current Practices and Considerations

    Developing new project: Predicting TPD Clinical Outcomes through PK/PD Assessment for Safer Therapies

    • Objective: To identify key drivers of translatability in exposure estimation and highlight data gaps that currently limit accurate predictions of clinical potency and safety margins. The ultimate goal is to develop a framework to better estimate clinical potency of TPDs based on nonclinical data.

  • QTc Challenges (New in 2026)

    This group is discussing and optimizing experimental approaches to better detect effects of TPDs on cardiac repolarization and predict QTc liability.

    New project underway: Evolving approaches to reduce the risk of delayed QTc prolongation with TPDs

    Objectives:

    • Develop or optimize study designs to ensure detection of potential atypical, delayed onset QT prolongation, ideally by adapting existing in vitro assays.
    • Develop and communicate a consensus framework of experimental methods that optimally detect effects of TPDs on cardiac repolarization to better predict QTc liability in humans.
    • Draw conclusions and propose hypotheses on the underlying mechanism of action based on shared data and proposed predictive models, if feasible.
